Genome editing to analyseressource allocation
Gene inactivation is a key step in studying bacterial functions and developing applications in synthetic biology. We used and improved a CRISPR-based genome editing technology to introduce mutations that stop the translation of targeted genes in E. coli.
This approach was deployed at the genome scale to inactivate the expression of a large number of genes, with the aim of freeing ribosomes from non-essential functions and reallocating them to functions of interest.
I will present the approach, the developments made to improve the system’s efficiency, and illustrate it with ongoing work that makes use of this genome editing tool.

Towardssustainableand autonomousluteinproduction – a heterotrophicmicroalgaeroute
The presentation will cover part of theresearch activities carried out at the Chair of Biotechnology of CentraleSupélec aimed at developing a biotechnologicalprocess for lutein production.
Lutein is a yellow carotenoid known for its beneficial effects on human health,particularly brain health. Historically supplied through green vegetables inthe diet, its intake has progressively decreased in modern eating habits(approximately 1.7 mg/day compared with the recommended 6-14 mg/day).
As a result, supplementation may become necessary. Current production processesare neither sustainable nor domestic. Our group is therefore developing of abiotechnology-based production route using microalgae, which can contain up to1000 times more lutein per serving than green vegetables.
After introducing this context, the presentation will be organized into threeparts. First, it will address an alternative to the conventional photosynthetesis: heterotrophic cultivation (bioprocessperspective). Second, it will explore (modestly) the physiological differencesbetween photoautotrophic and heterotrophic cells, including variations inlutein content. Finally, in a broader perspective, the numerical andcomputational tools supporting these investigations will be highlighted.
