Development of non-model recalcitrant ethanologenic bacterium Zymomonas mobilis as an efficient biorefinery chassis
Abstract
Efficient and robust microbial cell factories are the key for the burgeoning bioeconomy, which needs not only model microorganisms with rich information and abundant tools, but also non-model species with excellent industrial characteristics. Zymomonas mobilis is a natural ethanologen with many unique physiology characteristics making it an ideal industrial microbial biocatalyst. However, Z. mobilis faces the inherent limitations of inadequate knowledge, toolkits, and native product line that are usually associated with non-model microorganisms, which restrains the development of Z. mobilis as an efficient biorefinery chassis. I will present our efforts to overcome these challenges including the establishment of a one-stop centralized knowledgebase of ZymOmics (www.zymomics.cn), and our current effort to develop toolkits for synthetic biology practice including the development of efficient CRISPR-cas systems and the establishment of dual-reporter gene system for systematical identification and characterization of biological parts such as promoters, terminators, and 5’ UTR, as well as regulatory network by characterizing the global regulators and their interactions. I will also present our work to develop Z. mobilis for other bioproducts using lignocellulosic biomass such as 2,3-butanediol, isobutanol, PHB, and lactate. In addition, efforts of establishing a startup company for economic production of platform biochemicals using non-food feedstocks will also be discussed.
